The post received more than 101,000 views and 25,000 likes, with some of the account's 491,000 followers sharing their thoughts in the comment section.
De Rossi shared the same video clip on her personal Instagram account along with the hashtags "10 year anniversary" and "marriage equality."
DeGeneres and De Rossi met in 2004 but only married four years later when the ban on same-sex marriages was lifted.
The 45-year-old De Rossi wore a halter bridal gown designed by Zac Posen with a tulle skirt while DeGeneres wore a white pants suit and sheer Posen shirt.
The ceremony was held at their house, attended only by members from their inner circle. Part of De Rossi's vows was:
"It is good to be loved. It is profound to be understood."
Ellen added that her wife understands her, and even though it seems like something minor, it was "the best gift that anybody could've given me."
De Rossi legally changed her name to Portia Lee James DeGeneres, and the couple lives in California with their pets.
She left the film industry a few months ago to pursue other passions and has since opened a gallery called General Public. Read more about her career change here.
Fans of DeGeneres can look forward to her upcoming projects. She produced two made-for-TV films called "Couple Time" and "Little Funny." A release date hasn't been confirmed yet.
"Nancy Drew and the Hidden Staircase" is currently in post-production and will be released next year, while "Jekyll" is still in pre-production.
